Bells Line of Road is closed in both directions after a semi-trailer rolled at Mount Tomah about midday.
A Transport Management Centre spokeswoman advised motorists to use the Great Western Highway as an alternative route.
"Motorists already on Bells Line of Road are being turned around at the scene," said the spokeswoman.
"Emergency services are on site and the closure is expected to be in place for some time."
